Bio-Speedy® CRE Real-Time PCR Screening Kit’s principle is based on the steps that consist of:
1) Reactions are established by combining the “2X qPCR Mix” component, the “Target specific – Oligo Mix” component and the template DNA.
2) Positive results with internal control targeted oligonucleotides indicate that the reagents work, qPCR is not inhibited by exogenous template. The internal control target should be positive for all reactions in which the sample, PC and NC is added as a template.
3) Both CRE and Internal control targets should be positive in all reactions where PC is added as a template. PC template is used to confirm that CRE targeted oligonucleotides work.
4) The CRE target must be negative in all reactions where NC is added as a template. The NC template is used to confirm that there is no contamination that could react with the surrounding CRE targeted oligonucleotide.
5) Positive results with CRE-targeted oligonucleotides used as template for the sample in the reactions indicate that the sample is CRE positive.
INTENDED USE
Bio-Speedy® CRE Real-Time PCR Screening Kit is used to detect the DNA of Carbapenem Resistant Enterobacteriaceae (CRE) in DNA isolates obtained from clinical specimens, rectal swab, stool, blood, serum, CSF, etc. by real-time quantitative PCR (qPCR). In the scope of cre scans, blaKPC, blaNDM, blaVIM, blaIMP, blaOXA-48, blaOXA-51, blaOXA-23, blaOXA-58, blaTEM, blaSHV, bla(CTX-M), bla(CTX-M-15), bla(CTX-M-5) ve bla(CTX-M-3) genes are targeted by the kit
