The principle of the Bio-Speedy® MERS-CoV RT-qPCR Detection Kit consists of the steps:
Total RNA isolation from samples such as nasopharyngeal and / or oropharyngeal swab specimens, sputum *, saliva *, lower respiratory tract aspirate *, serum, faeces etc. is applied.
*Lower respiratory tract specimens are preferred for MERS-CoV detection. At different times, samples taken from different points increase diagnostic yield.
For qPCR MERS-CoV detection, multiplex RT-qPCR test with FEX stained labeled UPE and N2 target oligo sequences and HEX labeled Internal Control oligos is performed. In the HEX channel, the positive proliferation curve is examined and the internal status of the Internal Control is verified, then, if one or both of the upE and N2 targets are positive, a proliferation curve in the FAM channel is examined, and if Negative Control and Positive control are appropriate in the FAM channel, the MERS-CoV Positive result is recorded. Positive result validation is then performed by applying the N3 RT-qPCR test with FAM staining to the sample.

Bio-Speedy® MERS-CoV RT-qPCR Detection Kit is used for the detection of the virus called the Middle East Respiratory Syndrome Coronavirus (MERS-CoV) in clinical specimens for the diagnosis of Mers-CoV disease.
