1)Reactions of the Bio-Speedy® MRSA Real-Time PCR Screening Kit are established by combining the “2X qPCR Mix” component, the “MRSA – Oligo Mix” component and the template DNA.
2) Positive results with internal control (HEX) -targeted oligonucleotides indicate that the reagents work and qPCR is not inhibited by exogenous template. The internal control (HEX) target should be positive in all reactions to which the sample, the PC and NC are added as templates.
3) Both MRSA (FAM) and Internal control (HEX) targets should be positive in all reactions where PC is added as a template. PC template is used to confirm that MRSA (FAM) targeted oligonucleotides are working.
4) MRSA (FAM) target should be negative and internal control (HEX) target should be positive in all reactions where NC is added as a template. The NC template is used to confirm that there is no contamination that could react with the surrounding MRSA (FAM) targeted oligonucleotide.
5) Positive results with MRSA (FAM) -targeted oligonucleotides in the reactions used as templates for the sample indicate that the sample is MRSA positive.
INTENDED USE
Bio-Speedy® MRSA Real-Time PCR Screening Kit is used for detection of methicillin Resistant Staphylococcus aureus (MRSA) DNA from DNA isolates obtained from clinical specimens like Nasal swab, blood, serum, CSF etc. by real-time quantitative PCR (qPCR). The scope of the MRSA survey with the kit was targeted at the orfX-SCCmec intersection area.
