Bio-Speedy® Neisseria meningitidis Real- Time PCR Serotyping Kit’s serotype detection is based on the principle whose steps are:
1) The serotypes of B, C, W135, X and Y are identified by kit targeting Neisseria meningitidis bacterial strains genes “sacB”, “synD”, “synE”, “synG”, “synF” and “xcbB” respectively.
2) When the NmeA-Oligo Mix is applied in the clinical sample, the positive result observed in the FAM channel indicates that “N. meningitidis serotype A ” is positive in the sample; Positive result observed when NmeB-Oligo Mix was applied indicates that “N. meningitidis serotype B & quot is positive in the sample; Positive result observed when NmeC-Oligo Mix was applied indicates that “N. meningitidis serotype C ” is positive in the sample; The positive result observed when NmeW135-Oligo Mix was applied indicates that “N. meningitidis serotype W135 & quot is positive in the sample; Positive result observed when Nmex-Oligo Mix was applied indicates that “N. meningitidis serotype X & quot is positive in the sample; The positive result observed when applying NmeY-Oligo Mix indicates that “N. Meningitidis serotype Y “is positive in the sample. The negative result observed in the FAM channel indicates that these serotypes are not found in the sample.
3) When NmeA- Oligo Mix, NmeB- Oligo Mix, NmeC- Oligo Mix, NmeW135- Oligo Mix, NmeX- Oligo Mix and NmeY- Oligo Mix are applied to “NC” negative results indicate that there is no contamination.
4) Positive results (Ct ≤ 35) observed when the target-Oligo Mixes were applied to “PC Nme-SE” indicate that there is no loss of efficiency in PCR reagents and / or positive controls.
