1) Reactionsof the Bio-Speedy® VRE Real-Time PCR Screening Kit are established by combining the “2X qPCR Mix” component, the “VRE – Oligo Mix” component and the template DNA.
2) Positive results with internal control (HEX) -targeted oligonucleotides indicate that the reagents work and qPCR is not inhibited by exogenous template. The internal control (HEX) target should be positive for all reactions where the sample, the PC and NC is added as a template.
3) Both VRE (FAM) and Internal control (HEX) targets should be positive in all reactions where PC is added as a template. The PC template is used to confirm that the VRE (FAM) targeted oligonucleotides are working.
4) VRE (FAM) target should be negative and internal control (HEX) target should be positive in all reactions where NC is added as a template. The NC template is used to confirm that there is no contamination that could react with the surrounding VRE (FAM) targeted oligonucleotide.
5)Positive results with VRE (FAM) targeted oligonucleotides in the reactions which sample used as tamplate indicate that the sample is VRE positive.
INTENDED USE
Bio-Speedy® VRE Real-Time PCR Screening Kit uses for the detection of vancomycin-resistant Enterococci (VRE) via real-time quantitative PCR (qPCR) applied to DNA isolates obtained from clinical specimens like rectal swab, stool, blood, serum, BOS, etc. The vanA and vanB genes were targeted under the VRE screening kit.
